Abstract: Single-use systems have become an essential part of research and the production of biopharmaceuticals. The use of single-use systems (also called "disposables") offers a high level of biosafety and process reliability. Currently, one of the most common disposable bioreactors is a wave-mixed system. They are mainly used in cell culture technology for the cultivation of mammalian cells. The oxygen mass transfer coefficient is a limiting factor in wave-bag cultivation systems due to surface aeration. We therefore propose wave-mixed systems as a potential tool for the cultivation of strictly anaerobic microorganisms. In cooperation with PharmaBiome AG, a feasibility study for the cultivation of strictly anaerobic intestinal bacteria was carried out. The recording of the redox potential is essential for the process control of anaerobic cultivation. In addition, anaerobic inoculation methods have to be feasible, even outside the anaerobic workbench. Therefore, the modification of the wave-bag included the installation of a redox probe and an anaerobic inoculation method.
